Phosphate Buffered Saline (PBS)
Overview
Similar pH, electrolytes, and osmolarity to the human body.
- Concentration: 1x
- pH 7.4
Uses
- Washing tissues, cells
- Common base solvent in many solutions
Components
- \(NaCl\)
- \(KCl\)
- \(Na_2HPO_4\)
- \(KH_2PO_4\)
- \(diH_20\)
- 1L sterile container
Preparation for 1L of 1x PBS
- Add 800 mL of \(diH_20\) a sterile container
- Add electrolytes
- 8g \(NaCl\)
- 0.2g \(KCl\)
- 1.44g \(Na_2HPO_4\)
- 0.245g \(KH_2PO_4\)
- Adjust pH to 7.4 by titrating appropriate amounts of \(Na_2HPO_4\) or \(KH_2PO_4\)
Preparation for 1L of 10x PBS
- Add 800 mL of \(diH_20\) a sterile container
- Add electrolytes
- 80g \(NaCl\)
- 2g \(KCl\)
- 14.4g \(Na_2HPO_4\)
- 2.45g \(KH_2PO_4\)
- Adjust pH to 7.4 by titrating appropriate amounts of \(Na_2HPO_4\) or \(KH_2PO_4\)
